In animal and early human studies, ipamorelin produces pulsatile growth hormone release and a secondary rise in insulin-like growth factor 1. The magnitude and duration of that rise depend on route, sampling schedule, and the baseline endocrine state of the subject. Whether repeated exposure alters the response over time is not firmly settled, since some reports describe stable pulsatility while others note attenuation. Most published data come from small samples, which limits the strength of any general claim about long-term behavior.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class of compounds. Its sequence is Aib-His-D-2-Nal-D-Phe-Lys-NH2, a structure that differs from natural ghrelin in length and in the presence of non-natural amino acid residues. Early laboratory work described it as a comparatively selective agent that stimulates growth hormone release with limited effect on other pituitary hormones. The compound is supplied as a lyophilized solid for research use and has no identified natural source in the body.
Human data remain limited and come mainly from small, short-term studies conducted decades ago. The peptide has not received approval as a medicine from major regulators, so current availability is largely as a research chemical. Reported effects on growth hormone pulsatility, appetite, and body composition should be read as preliminary, since few independent groups have replicated the original findings. Analytical characterization of research-grade material varies between suppliers, which complicates comparison across studies. Regulatory status also differs by country, and some jurisdictions classify it as a prescription-only or otherwise restricted item.
Ipamorelin is a synthetic pentapeptide that belongs to the growth hormone secretagogue class. It acts as an agonist at the ghrelin receptor, also called the growth hormone secretagogue receptor type 1a. The compound was designed in the 1990s during a search for agents that release growth hormone with fewer off-target hormonal effects than earlier secretagogues. It appears in the research literature under several sequence-based names. Material supplied for laboratory work is normally a lyophilized solid, and it is not marketed as an approved therapeutic in major jurisdictions.

One distinguishing feature reported in animal studies is selectivity. Ipamorelin stimulated growth hormone release with limited elevation of adrenocorticotropic hormone or cortisol compared with earlier secretagogues such as GHRP-6. This pattern has been described as more selective for the growth hormone axis. The finding comes mainly from preclinical work, and the degree to which it holds across species and doses is not fully settled. Reports also describe effects on gastric motility in animal models, suggesting activity outside the pituitary, though the clinical relevance of this observation is uncertain.
Ipamorelin is a synthetic pentapeptide that acts on the growth hormone secretagogue receptor, also known as the ghrelin receptor. Its sequence contains five amino acid residues, including a non-natural residue that increases stability against enzymatic breakdown. The compound was developed in the 1990s as part of research into small peptides that stimulate pituitary hormone release. Unlike larger protein hormones, it can be produced by solid-phase peptide synthesis and characterized by standard analytical methods.
At the receptor level, ipamorelin binds GHS-R1a and triggers signaling through Gq-coupled pathways. Activation leads to calcium release and downstream effects in pituitary somatotroph cells. These events promote the release of growth hormone into circulation. The response depends on the presence of the receptor and on the physiological state of the animal or tissue studied. Because the receptor is also found in other tissues, effects beyond the pituitary have been examined in laboratory models, though the extent of those effects remains an area of ongoing study.
Structural features distinguish the molecule from earlier secretagogues. An alpha-aminoisobutyric acid residue near the N-terminus and a D-naphthylalanine substitution increase receptor affinity, while C-terminal amidation improves resistance to exopeptidases. These modifications are associated with reduced stimulation of appetite and of the hypothalamic-pituitary-adrenal axis compared with hexarelin or growth hormone releasing peptide-6. Whether the same profile applies at every dose level studied is a matter of ongoing investigation rather than settled consensus.
Ipamorelin is a synthetic pentapeptide that acts as an agonist at the ghrelin receptor, also called the growth hormone secretagogue receptor type 1a. Its sequence incorporates non-natural residues, which slows enzymatic breakdown relative to short native peptides. In laboratory and early clinical work the compound is described as a selective growth hormone secretagogue because it raises growth hormone with comparatively little effect on other pituitary outputs. The degree to which that selectivity holds across species and dosing regimens remains an open question in the published literature.
Signal transduction begins when the peptide binds GHSR-1a on pituitary somatotrophs. The receptor couples to Gq/11 proteins, activating phospholipase C, which cleaves phosphatidylinositol bisphosphate into inositol trisphosphate and diacylglycerol. Inositol trisphosphate releases calcium from intracellular stores, and the resulting rise in cytosolic calcium drives growth hormone vesicle fusion. Concurrent Gs coupling and cyclic AMP elevation have also been reported, and the relative contribution of each arm to the overall secretory response is not fully settled.
Long-term storage of the dry powder is typically at minus twenty degrees Celsius or lower, protected from light and moisture. Solutions are commonly kept frozen and thawed only once, because repeated freeze-thaw cycles can promote aggregation and loss of measurable peptide content. Buffers near neutral pH are preferred over strongly acidic or strongly basic conditions. Shipping at ambient temperature is acceptable for short periods when the powder remains sealed and desiccated.
Identity and purity are assessed by complementary methods rather than a single test. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and reports a percentage purity. Mass spectrometry, most often with electrospray ionization, confirms the expected molecular mass and detects sequence-related variants. Amino acid analysis can verify composition, while water content and residual counterion measurements support the mass balance of a batch. Stability studies under accelerated conditions are used to estimate shelf life, though such estimates carry uncertainty for long-term storage.
Material supplied for research use is normally a white to off-white lyophilized powder. The solid is hygroscopic and is handled in a low-humidity environment to limit water uptake. Bulk quantities are frequently shipped in sealed vials under inert gas. Once reconstituted in water or a neutral buffer, the solution is less stable than the dry powder and is usually divided into single-use aliquots.

=== Charter schools === Moore does not support the expansion of charter schools, saying that he wants to focus on improving public school districts, but wants to ensure accountability for current charters. In his first budget, in January 2023, Moore cut funding for the state's Broadening Options and Opportunities for Students Today (BOOST) program to provide scholarships to students attending charter schools by $2 million, and introduced new eligibility limits for current BOOST students and their siblings. In an interview with Jewish Insider, Moore said he intended to get rid of the BOOST program in a few years, adding, "The focus that I have, the focus that our administration is going to have, is making sure that we are creating and developing world-class public schools for our students." The budget was amended to reduce these cuts by $1 million in March 2023, and signed into law in April 2023. Moore maintained funding for the BOOST program in his 2024 budget.
=== Elderly === Skin ageing is associated with increased skin vulnerability, and the texture and colour of the skin can change over time. Although wrinkles occur naturally due to ageing, smoking can intensify the appearance of wrinkles. Sunspots, dryness, wrinkles, and melanomas can occur from UV exposure over time, whether it be through the sun or through tanning beds. Exposure to UV can make skin less elastic. Skin problems including pruritus are common in the elderly but are often inadequately addressed. A literature review of studies assessing the maintenance of skin integrity in the elderly found most studies to have low levels of evidence, but the review concluded that skin-cleansing with synthetic detergents or amphoteric surfactants induced less skin dryness than using soap and water. Moisturizers with humectants helped with skin dryness and skin barrier occlusive reduced skin injuries. When taking baths or showers, using warm water rather than hot water can aid with dryness.
Tim Hortons, currently owned by Restaurant Brands International, opened its first store on May 17, 1964, in Hamilton, Ontario. It is the fourth largest publicly traded quick-service restaurant chain in North America based on market capitalization, and the largest in Canada. It has cornered the Canadian market for baked goods (76%) and coffee (62% compared to Starbucks, in the number two position, at 7%). The company has experienced substantial growth in the United States, where it has 613 stores (April 2011, about 220 more than it had in June 2008). It has a same-store sales growth rate of 3.9% in Canada and 4.9% in the US. The new partnership with Cold Stone Creamery helped push its US presence to over 600 stores. The partnership has since dissolved. Sales exceeded $2 billion in 2008. It was controlled by Wendy's International Inc. for 11 of the 46 years of operation, from August 8, 1995, until the end of 2006. On April 3, 2011, Tim Hortons oversaw 3,782 locations in North America 3,169 of which were in Canada. In the fourth quarter of 2010 Tim Hortons' profits were 27% more than they were over the entire 2009 fiscal year (for the quarter net income was up 314%). In Forbes' 2011 edition of the world's 2000 largest companies, Tim Hortons placed 1469th overall (up from 1714th in 2010) with annual sales of $2.6 billion (up from $2.014 billion in 2010), profits totalling $628 million (up from $280 million), assets worth $2.5 billion (up from $1.90 billion) and a market value of $7.5 billion (up from $5.41 billion).
== Uses == As of 2022, alginate had become one of the most preferred materials as an abundant natural biopolymer. It is particularly useful as a biomaterial because of its nontoxicity, hygroscopicity, and biocompatibility, and can imitate local bioenvironments; its degradation product can be easily cleared by the kidneys. Alginate absorbs water quickly, which makes it useful as an additive in dehydrated products such as slimming aids, and in the manufacture of paper and textiles. Alginate is also used for waterproofing and fireproofing fabrics, in the food industry as a thickening agent for drinks, ice cream, cosmetics, as a gelling agent for jellies, known by the code E401 and sausage casing. Sodium alginate is mixed with soybean protein to make meat analogue. Alginate is used as an ingredient in various pharmaceutical preparations, such as Gaviscon, in which it combines with bicarbonate to inhibit gastroesophageal reflux. Sodium alginate is used as an impression-making material in dentistry, prosthetics, lifecasting, and for creating positives for small-scale casting. Sodium alginate is used in reactive dye printing and as a thickener for reactive dyes in textile screen-printing. Alginates do not react with these dyes and wash out easily, unlike starch-based thickeners. It also serves as a material for micro-encapsulation.

==== Education and intelligence ==== Research suggests neither a good education nor a high IQ reliably increases happiness. Anders Ericsson argued an IQ above 120 has a decreasing influence on success. Presumably, IQs above 120 do not additionally cause other happiness indicators like success (with the exception of careers like Theoretical physics, where high IQs are more predictive of success). Above that IQ level, other factors, like social skills and a good mentor, matter more. As these relate to happiness, intelligence and education may simply allow one to reach a middle-class level of need satisfaction (as mentioned above, being richer than this seems to hardly affect happiness). According to the findings of the study, Using Theatrical Concepts for Role-plays with Educational Agents by Klesen, she expresses how role- playing embeds information and educational goals and causes people to learn unintentionally. Studies have shown that enjoyment in things as simple as role playing increases a person's IQ and their happiness. Martin Seligman has said: "As a professor, I don't like this, but the cerebral virtues—curiosity, love of learning—are less strongly tied to happiness than interpersonal virtues like kindness, gratitude and capacity for love."
Arguably its biggest change to date, in 1959 Guinness began using nitrogen, which changed the fundamental texture and flavour of the Guinness of the past as nitrogen bubbles are much smaller than CO2, giving a "creamier" and "smoother" consistency over a sharper and traditional CO2 taste. This step was taken after Michael Ash—a mathematician turned brewer—discovered the mechanism to make this possible. Nitrogen is less soluble than carbon dioxide, which allows the beer to be put under high pressure without making it fizzy. High pressure of the dissolved gas is required to enable very small bubbles to be formed by forcing the draught beer through fine holes in a plate in the tap, which causes the characteristic "surge" (the widget in cans and bottles achieves the same effect). This "widget" is a small plastic ball containing the nitrogen. The perceived smoothness of draught Guinness is due to its low level of carbon dioxide and the creaminess of the head caused by the very fine bubbles that arise from the use of nitrogen and the dispensing method described above. Foreign Extra Stout contains more carbon dioxide, causing a more acidic taste. Although Guinness is black, and is referred to as "the black stuff" in Diageo marketing, it is also "officially" referred to as a very dark shade of ruby. The most recent change in alcohol content from the Import Stout to the Extra Stout was due to a change in distribution through North American market. Consumer complaints influenced subsequent distribution and bottle changes.
This is a list of radioactive nuclides (sometimes also called isotopes), ordered by half-life from shortest to longest, in seconds, minutes, hours, days and years. Current methods make it difficult to measure half-lives between approximately 10−19 and 10−10 seconds.
δDs of annual precipitation in different regions of the world are shown on the map. The precipitation is more D-enriched near the equator in the Tropics. The δDs generally fall in the range of around −30 ~ −150‰ in the northern hemisphere and −30~+30‰ over land areas of the southern hemisphere. In North America, the δD of average monthly precipitation across regions is lower in January (ranging up to around −300‰ in Canada) than in July (up to around −190‰). The overall mean precipitation is determined by the balance between evaporation of water from the oceans and other surface water and condensation of water vapor in the form of rain. Net evaporation should equal net precipitation, and the δD for precipitation is around −22‰ (global average). The Global Network of Isotopes in Precipitation (GNIP) investigates and monitors the isotopic composition of precipitation at various sites all over the world. The mean precipitation can be estimated by the equation, δ2H = 8.17(±0.07) δ18O + 11.27(±0.65)‰ VSMOW. (Rozanski et al., 1993) This equation is the slightly modified version from the general global meteoric water line (GMWL) equation, δ2H = 8.13δ18O + 10.8, which provides the average relationship between δ2H and δ18O of natural terrestrial waters.

It is possible that it may be related to Cirroteuthis, and in that case the two posterior stumps, looking like arms, may be the remains of the lateral fins, for they seem too far back for the arms, unless pulled out of position. On the other hand, they seem to be too far forward for fins. So that they are probably arms twisted out of their true position. However, having examined samples of the mass sent to him by Webb, Verrill concluded that "the creature cannot be an Octopus, but is of cetacean nature." He suggested that "the whole mass represents the upper part of the head of [a sperm whale], detached from the skull and jaw." Webb decided the carcass should be moved further inland so that it would not be lost to the sea forever. With the help of "six horses and strong tackle", it was moved several miles closer to St. Augustine, "to the terminus of a railroad," where it was protected from the tide and drifting sand. Its final resting place was South Beach, Anastasia Island, near the hotel of Dr. George Grant. The St. Augustine carcass became somewhat of a tourist attraction and was visited by large numbers of people. It is unknown what happened to the carcass afterwards. Photographs of the St. Augustine carcass were for a long time thought to be lost, and drawings remained the only pictorial evidence of the event. They were finally rediscovered in 1993 by Marjorie Blakoner of California, who recognised them in an old album. Van Lockwood, one of the original photographers of the St. Augustine carcass, kept an album of photos he had taken between 1885 and 1899.
Mark Salisbury of Time Out wrote of the lead acting to be "excellent" and the crowd scenes "astonishing", while equally observing how the climax was "truly nerve-wracking". He called it "an implacable work of authority and compassion, Cry Freedom is political cinema at its best." James Sanford, however, writing for the Kalamazoo Gazette, did not appreciate the film's qualities, calling it "a Hollywood whitewashing of a potentially explosive story." Rating the film with 3 Stars, critic Leonard Maltin wrote that the film was a "sweeping and compassionate film". He did, however, note that the film "loses momentum as it spends too much time on Kline and his family's escape from South Africa". But in positive followup, he pointed out that it "cannily injects flashbacks of Biko to steer it back on course." John Simon of the National Review called Cry Freedom "grandiosely inept". In 2013, the movie was one of several discussed by David Sirota in Salon in an article concerning white saviour narratives in film.

The AH50 assay is based on the lysis of unsensitized rabbit erythrocytes, however, normal results have been reported in patients with Type I properdin deficiency. Family history of X-linked inheritance should be considered. Further testing includes: Factor D Function by Hemolytic Assay, Properdin Level by ELISA, and gene sequencing to detect mutations for confirmation. Patients with Factor D deficiency or properdin deficiency are advised to receive meningococcal vaccinations and frequent evaluations for meningococcal antibodies. For those with recurring infections, prophylactic antibiotics are administered.
is referred to as the chi plot. For flat surfaces, the slope of the chi plot yields the surface area. Empirically, this plot was noticed as being a very good fit to the isotherm by Michael Polanyi and also by Jan Hendrik de Boer and Cornelis Zwikker but not pursued. This was due to criticism in the former case by Albert Einstein and in the latter case by Brunauer. This flat surface equation may be used as a "standard curve" in the normal tradition of comparison curves, with the exception that the porous sample's early portion of the plot of
=== 3 May === French president Emmanuel Macron announced he would consider sending French troops to Ukraine "if the Russians were to break through the front lines" or if Ukraine were to request it. In Donetsk Oblast, three people were killed by Russian attacks in Kurakhove and Chasiv Yar, while one person was killed in a separate attack in Kharkiv. In Russia, the governor of Belgorod Oblast claimed that two people were injured by Ukrainian drone strikes in Voznesenovka and Shebekino. The HUR claimed the destruction of two Russian locomotives by fire over the last few days: one in Orenburg, the other in Vladikavkaz. Ukrainian media also reported that the agency carried out a cyberattack on internet providers and mobile operators in Tatarstan. The SBU arrested a resident of Kharkiv on suspicion of spying for Russia. The FSB claimed to have "liquidated" an agent working for the HUR who was plotting to carry out terrorist attacks against "military and energy objects" in Moscow and Leningrad Oblasts. It is a synthetic pentapeptide in the growth hormone secretagogue family and acts as an agonist at the ghrelin receptor. It is handled as a laboratory research compound rather than a naturally occurring hormone.
Studies generally report growth hormone as the dominant output, with smaller or absent effects on adrenocorticotropic hormone and prolactin. The size of that separation depends on the assay and the dose examined, so it is best described as relative selectivity.
No natural source has been identified, and the molecule is produced by chemical synthesis. Its non-natural residues distinguish it from endogenous ghrelin even though both engage the same receptor.
It has not been approved as a therapeutic by major regulators, and the human trial record is small and dated. Material available today is mostly sold as a research chemical for laboratory use. Approval and restriction status varies by country.
